DR DAVID BARNETT

Biography | Research Interests | Publications

Dr. Barnett’s research is primarily in the area of analytical mass spectrometry (MS), and its application to the identification and characterization of biological macromolecules, focusing on proteins and peptides. We are particularly interested in applying various analytical techniques, centred on MS detection, to the investigation of protein complexes, protein-protein interaction networks, protein isoforms, post-translational modifications and relative protein expression between normal and diseased biological samples.

In addition to establishing a mass spectrometer based platform for high-throughput protein identification at the Institute, David’s work will entail development and/or application of novel, state-of-the-art, sample preparation and fractionation techniques as well as multi-dimensional separations aimed at reducing the complexity of the human proteome.
 
Because of the complex nature of the field of “proteomics”, energy will also be focused on the optimization of a differential protein labelling methodology for relative expression studies and advanced data interpretation and analysis with external collaborators.(( Dr. Barnett will also use his instrumental expertise to continue to develop FAIMS technology for rapid gas-phase fractionation of protein samples to improve the overall peak capacity and detection capability of conventional multi-dimensional chromatographic separations coupled to mass spectrometry.
 
The characterization of protein tertiary and quaternary structure (intact proteins) using FAIMS technology will also be pursued as a probe of protein folding or mis-folding and specific protein-protein and protein-ligand interactions, respectively.
